Magic Media Marker has the ability to strip video out of an mkv container, convert the audio if needed, and put it all back together in a m4v container that Apple's devices can read. This is called remuxing, not only does it save time over converting files, but it also avoids any quality loss since it is not transcoding the video. Magic Media Marker 's main purpose is to process video files and make them pretty in iTunes and on your Apple Devices. Feed Magic Media Marker a video file and it will automatically determine if the file is a TV Show or a Movie by it's file name and download the appropriate meta data and artwork for you. Windows Movie Maker 2012 works fine on Windows 10, Windows 8/8.1 and Windows 7. Officially, Microsoft has stopped supplying Windows Essentials 2012 (the last release). For a while you could still downloaded the small installer (wlsetup-web.exe 1.2MB) from Microsoft.com but that app can’t grab the larger install files.

  • thialysine (50mg/ml)
  • clonNat (50mg/ml)
  • G418 (142 mg/ml)
  • hygromycin (50mg/ml)

For 1L of media the final composition is:

  • 1.7g YNB without amino acids
  • 1 g Monosodium glutamate
  • 20g bacto-agar (2% bacto-agar)
  • 20g glucose (2% glucose)
  • canavanine (50mg/L final)
  • thialysine (50mg/L final)
  • clonNat (50mg/L final)
  • G418 (142mg/L final)
  • hygromycin (50mg/L final)

Procedure

Combine:

Billboards
  • 1.7g YNB w/o amino acids or ammonium sulfate
  • 1g monosodium glutamate
  • 20g bacto-agar

Bring volume to 900ml with mili-Q water. Autoclave.

*Megan N McClean: Every time you do this protocol you need to check the drug stock concentrations. For example, the last time the G418 stock was made up the activity of the drug wasn't taken into account, which is why it is a 142mg/ml stock instead of a 200mg/ml stock. Double-check the stock solutions and your final desired concentration before making up the media.
